Researchers searching for a cure for obesity said on Thursday they have developed a drug that not only makes mice lose weight, but reverses diabetes and lowers their cholesterol, too.

The drug, which they have dubbed fatostatin, stops the body from making fat, instead releasing the energy from food. They hope it may lead to a pill that would fight obesity, diabetes and cholesterol, all at once.

Writing in the journal Chemistry and Biology, Salih Wakil of Baylor College of Medicine in Texas, Motonari Uesugi of Kyoto University in Japan and colleagues said the drug interferes with a suite of genes turned on by overeating.

“Here, we are tackling the basics,” Wakil said in a telephone interview. “I think that is what excited us.”

Scientists are painfully aware that drugs that can make mice thin do nothing of the sort in humans. A hormone called leptin can make rats and mice drop weight almost miraculously but does little or nothing for an obese person, for instance.

But Wakil, whose team has patented the drug and is looking for a drug company to partner with, hopes this drug may be different. “I am very, very optimistic,” he said.

Fatostatin is a small molecule, meaning it has the potential to be absorbed in pill form.

It works on so-called sterol regulatory element binding proteins or SREBPs, which are transcription factors that activate genes involved in making cholesterol and fatty acids.

“Fatostatin blocked increases in body weight, blood glucose, and hepatic (liver) fat accumulation in (genetically) obese mice, even under uncontrolled food intake,” the researchers wrote.

Genetic tests showed the drug affected 63 different genes.

The idea of interfering with SREBP is not new. GlaxSmithKline has been working on a new-generation cholesterol drug that uses this pathway.

After four weeks, mice injected with fatostatin weighed 12 percent less and had 70 percent lower blood sugar levels, the researchers wrote.

Now they plan to test rats and rabbits, Wakil said.

The drug also had effects on prostate cancer cells they said — something that may help explain links between prostate cancer and obesity.

Cory Doctorow starts his article (Writing in the Age of Distraction) by acknowledging that his readers are overwhelmed by the myriad distractions that lie one click away on the Internet. Busted. I actually was reading his article as a deliberate distraction from my usual work.  Apparently he knows what he’s talking about. 

Cory shared that he’s able to manage his bent for distraction and keep on top of his workload by depending on a few techniques, one of which he introduced as a “short, regular work schedule.” He sets aside a specific amount of time to focus on what he is writing, every day, no matter what. The result is a body of work containing many books, columns, and blog posts every year.

What’s the relationship between writing and this weight loss journey?

For myself, I know that most of the time I am distracted and overwhelmed by the immensity of the weight loss challenge I see before me. If I had only a few pounds to lose, then maybe I could see the end of the dreaded diet up ahead somewhere. However, from where I stand now, I cannot see the end of it. Every time I think about losing weight, I also have to think about how in the world I am ever going to keep my momentum and dedication revved-up for long enough to see this task to its end.

What’s the dieter’s equivalent of writing for twenty minutes every day?

You could make yourself create a menu and shop for just one healthy, slimming meal per day.  Or, you could make yourself do just ten minutes of cardio work or strength training each day.  You could even just read my daily post and implement a single related change in your life.

That way, you could focus on your intentions and your behaviors, rather than on your outcome which might be good or bad, or even worse, result in no change at all.  You could have a go at accumulating enough good days to pile up a real difference in the same way that a writer builds up enough 20-minute sessions to accomplish the drafting of an entire book.

Human Growth Hormone (HGH)

HGH, otherwise known as the “Master Hormone,” can effectively fight the aging process. It controls the release of many other hormones in the body. As it turns out, some of my friends think about fat, and getting rid of it, nearly as much as I do.  We remind me of that television commercial where slightly pudgy women go through their days with bathroom scales chained to their ankles.

While downing a cheesy, spicy plate of manicotti the other evening, one of my dinner companions began describing her lunch time get-healthy habits.  She cheerfully offered that she had recently adopted the habit of munching a snack-sized bag of carrots every day because “carrots speed up your metabolism.”

(Sounds of my weight-loss-strategies-alarm ringing loudly)           

Well, now!  I just had to go do some research about that.

According to nutritionists, there truly are foods that help to boost your metabolism.  Generally, these foods are classified as complex carbohydrates, and they can help you in at least these three ways:

  • thermic effect — These foods require relatively more energy to process and use than other foods.  Some folks even go so far as to call them “negative calorie” foods, meaning they actually take more calories to digest than they contribute to your diet.

What:  (Trick for Lowered Cholesterol):  One Alli pill at each meal that contains more than a few grams of fat.

Why:  Alli blocks some dietary fat, causing it to pass from the body undigested.  One Alli can reduce the number of calories digested at every meal.

Research has shown that exercising in water can be a challenging and effective way to get fit and lose weight.  For me, exercising in water is a sure-fire way to feel stronger, more limber, and lots lighter.  Buoyancy allows me to ease into deeper stretches and less difficult strength-building moves, e.g., squats and push-ups, than I could manage if I were to attempt the same moves on land.

Aquatic exercise has been around for a long time.  It was originally used as a form of rehabilitative therapy for injured athletes.  Sports medicine experts discovered water’s buoyancy, resistance (12 times that of land), and therapeutic massaging effect contributed to the healing process.

Water acts as a giant cushion.  It saves wear and tear on your muscles and protects your joints.  As an added benefit, water pressure massages your muscles.

Water work outs are great ways to treat angry knees and ankles.  If you are submerged to your neck, gravitational pull on your body is reduced up to 90%.  Water training is also wonderful for developing overall balance and coordination.

Besides all that, it just feels good.

New Weight Loss Drug – Contrave

Contrave, a weight loss drug developed by Orexigen Therapeutics has completed its Phase III trial, and Orexigen is now seeking approval from the Food and Drug Administration.

The Phase III trial involved several thousand patients, and built on an earlier trial involving approximately 700 people.  In these trials, individuals receiving Contrave lost an average of 6.1% and 6.4% of their weight, respectively.  This result compared with 1.3% and 1/2%, respectively, for those taking a placebo.  In a third trial, obese patients with Type 2 diabetes showed similar weight losses.

Very few side effects were reported and primarily consisted of headaches and constipation.

Contrave is a combination of two other drugs:

Naltrexone:  Naltrexone is a medication that blocks the effects of drugs known as opioids (a class that includes morphine, heroin or codeine). It competes with these drugs for opioid receptors in the brain. It was originally used to treat dependence on opioid drugs but has recently been approved by the FDA as treatment for alcoholism. In clinical trials evaluating the effectiveness of naltrexone, patients who received naltrexone were twice as successful in remaining abstinent and in avoiding relapse as patients who received placebo-an inactive pill. 

In the largest study, the most common side effect of naltrexone affected only a small minority of people and included the following: nausea (10%), headache (7%), dizziness (4%), fatigue (4%), insomnia (3%), anxiety (2%), and sleepiness (2%).

Bupropion:  Bupropion (Wellbutrin) is used to treat major depressive disorder and seasonal affective disorder.  At least one brand (Zyban) is used to help people stop smoking by reducing cravings and other withdrawal effects.  The most common side effects of bupropion included:

Agitation — in up to 31.9 percent of people, Weight loss — up to 28 percent of people, Dry mouth — up to 27.6 percent of people, Constipation – up to 26 percent of people, Headaches – up to 25.7 percent of people, Nausea or vomiting — up to 22.9 percent of people.

Both Naltrexone and Bupropion are well-established and Orexigen Therapeutics reported no adverse psychological events in the Contrave trial.  However, it is worth noting that black box warnings were recently added to Zyban to alert consumers that taking the drug might cause hostility, depression and other mood changes.  Similar warnings have been noted for Wellbutrin as well.

On my weight loss journey, I am hopefully searching for any little aide to tip the scales in my favor, so to speak.  However, in my opinion, a 6% weight loss possibility seems like a small benefit compared to the risk of “hostility, depression, and other mood changes.”  I think I’ll pass on this drug when/if it becomes available.

When it comes to losing fat and building muscle, eating less food is not the answer. Considering impacts to your metabolism may be. 

Others will tell you that losing excess fat is simply a matter of using more calories than you eat. They sometimes forget to mention that eating less will slow your metabolism.

Part of the secret to losing weight by eating the right amounts of food while keeping your fat levels in check. is to use the thermic effect of food to your advantage.

The thermic effect is defined as the incremental energy requirement above your resting metabolic rate used due to the cost of digestion, absorption, and disposal of ingested food. 

Translation: Some of the foods you eat speed up your metabolism more than other foods.

You’ll find it much easier to reduce your fat levels if you consume plenty of foods with a higher thermic effect. Proteins tend to have a much higher thermic effect than other types of foods.

Calculating the thermic effect: A general guideline used by some to calculate the thermic effect of the foods you eat is to take your total calorie consumption and multiply that by 10% to get the total number of calories for the thermic effect. This method is a general estimation, and the thermic effect for different food types can range from 3-30%.

In general:

Fats: Thermic effect of about 3%. Keeping certain levels of fats in your diet is necessary, but fats tend to be high in calories and have a low thermic effect.

Fibrous vegetables: Thermic effect of about 20%. Many fruits and vegetables are negative calorie foods. Get a good portion of vegetable in at least half of your daily meals.

Proteins: Thermic effect of about 30%. High protein foods are essential for muscle gain and fat loss. Think of these foods as your metabolic stimulator. Have one portion with each of your meals.

Feel Better Fast – Avoid Overloading Yourself.

How is your weight loss project coming along? Losing weight? Feeling stronger, healthier, leaner? Me neither. I keep making new decisions and plans, and keep on doing the same old things that got me this way in the first place.

Sometimes, our best efforts at analysis, and planning, and hoping just don’t result in the slimmer person we just know we deserve to be.

In order to see a change in our bodies, we must effect a corresponding change in our actions. Believe me! You cannot wish away the padding around your waist. I’ve tried!  If what you are doing now is not resulting in what you want, then you have to change what you are doing. You must adjust. You must adapt and overcome (as my young daughter tells me).

Consider solution-oriented behavior modification. Think of one small change you can make for the better. Just one. Then do it every time you see the opportunity.

For example, you could observe that the double hamburger and fries that you have most days for lunch is contributing too many calories to your diet, and too much bad fat as well. Then, you could decide to make your “one change” at lunch. Tomorrow, instead of burger and fries, order a grilled chicken sandwich and fries instead. Not perfect for sure, but a step in the right direction. Now, instead of consistently having too many calories and too much fat for lunch every day, you are having lean protein and less fat. You can feel immediate satisfaction from your positive step, and over time, you will feel the positive effects on your health.  Pile one good change on top of another.
